MECHANISM OF ACTION
Dapoxetine is a selective serotonin reuptake inhibitor. It inhibits neuronal uptake of serotonin and increases the activity of serotonin at pre and postsynaptic receptors. Serotonin is a neurotransmitter responsible for ejaculation. Central nervous system regulates ejaculation in males. After stimuli from sex organs, spinal reflex originates ejaculatory pathway. These signals are transmitted to the brain. Studies showed that administration of dapoxetine inhibits ejaculatory expulsion reflex. Due to this time of ejaculation increases.
WHEN TO USE PRILIGY
PRILIGY is indicated for the treatment of premature ejaculation (PE) in males of 18 to 64 years of age who has:
- No control over ejaculation.
- Ejaculation without any stimulation or have ejaculation in less than 2 minutes after penetration, on most occasions
- Distress and interpersonal difficulty because of premature ejaculation.
What is the recommended dose?
PRILIGY is a drug which is to be used only when required; so it has to be taken 1-3 hours prior to engage in sexual activity. PRILIGY tablets are available in two strengths: 30 mg and 60 mg. Always start with the 30 mg tablet in a day, there should be a minimum gap of 24 hours between two doses. Do not start with a 60 mg dose: in case you are not getting any benefit from 30 mg dose tell your doctor and use the dose prescribed by him.
How to take PRILIGY?
PRILIGY has a bitter taste, so always take it with water. Do not crush or chew the tablet. It can be taken with or without food.
What are the side effects?
- Nervous system: change in taste, dizziness, fainting (syncope), decreased consciousness
- Gastrointestinal: vomiting, diarrhoea, constipation, abdominal upset, pain in stomach,
- Eyes related: blurred vision
- Heart related: slow and/or fast heart beat
- Ear related: vertigo
- Respiratory system: congestion
- Blood related: high or low blood pressure
- Skin related: pruritis, cold sweat
- Others: fatigue, irritation
CONTRAINDICATIONS
PRILIGY should not be administered:
- If you have allergy to dapoxetine or any other constituent or PRILIGY (always read labels before using any medicine)
- If you have cardio logical problems like heart failure; conduction problems (AV block, sinus syndrome); disease of the valve.
- If you have used MAO inhibitors (drugs for depression within the past 14 days)
- if you are using recreational drugs like LSD, narcotics, or benzodiazepines
- If have any problems related to the kidneys, liver and blood.
- If your age is above 65 years
- If you are a woman ( PRILIGY is meant for males only)
- If your age is below 18 years.
- If you have epilepsy (seizures).
- If you have lactose intolerance.
Always discuss your medical history with doctor before using any medicine.
What are the precautions?
- PRILIGY should be used only in males diagnosed with Premature Ejaculation. Always use it on prescription of a doctor.
- If you have erectile dysfunction and are using PDE5 inhibitors then always consult a doctor before using PRILIGY.
- If you have blood pressure problem then always take it under medical supervision.
- Do not use it if you are taking medicines for depression (MAO inhibitors).
- You should avoid driving or doing a work that requires alertness as PRILIGY cause drowsiness and light headedness.
- It is advised not to use alcohol with PRILIGY; it can lead to syncope (fainting).
Drugs that interact with PRILIGY
There are some drugs that interact with PRILIGY.
- Monoamine oxidase inhibitors (MAO)
- Thioridazine
- CYP3A4 inhibitors like itraconazole, ritonavir, saquinavir, nelfinavir, atazanavir, telithromycin, nefazodone.
- CYP2D6 inhibitors
- PDE 5 inhibitors like verdenafil, sildenafil, tadalafil.
- Warfarin
Always keep a list of prescription and non-prescription drugs and discuss it with your doctor before using PILIGRA.
What to do in case of overdose?
In case of overdose, immediately contact your nearest emergency centre.
STORAGE CONDITIONS
It should be stored below 25 degree Celsius in a dry place. Keep away from the reach of children.
